Mutations in the CRYAB gene cause distinct clinical phenotypes including isolated posterior polar cataract, myofibrillar myopathy, cardiomyopathy, or a multisystemic disorder combining all these features. Impairment of alpha-B crystallin dimerization may be relevant to the pathogenesis of these disorders.
